棉花中水溶性总糖含量的提取及测定方法研究

Extraction and Determination of Total Water-soluble Sugars in Cotton

摘要: 通过对蒽酮比色法测定棉花中水溶性总糖含量的几个问题进行了探讨,优化了蒽酮比色法测定棉花中水溶性总糖含量的反应条件及棉花中水溶性总糖的提取条件。 结果表明, 蒽酮比色法测定棉花水溶性总糖的适宜测定条件为:波长575 nm,显色剂添加量5 ml,显色时间10 min,静置时间10 min~1h均可,在此条件下葡萄糖标准品浓度与吸光度之间具有良好的线性关系(C= 0.131 8A+0.000 4,R2=0.999 2)。采用此优化条件测定棉花中水溶性总糖含量,灵敏度高(可达到0.003 mg.mL-1的检出限),具有良好的精密度(重复性标准偏差为0.04)和准确性(平均回收率为100.3%)。该测定方法简单、经济、快捷、准确,特别适合大批量样品,尤其是资金有限的实验室。

Abstract: The reaction condition and extracting condition of the anthrone colorimetric method to determine the soluble sugar content of cotton were investigated in this paper. The results showed that the optimum determination condition was: detector wavelength of 575 nm, color- display reagent consumption of 5 mL, reaction time of 10 min, reaction stalling time of 10 min to 1 h. Under this condition, there was a good linear relationship between the concentration of glucose and absorbance (C= 0.1318A + 0.0004, R2=0.9992). The limit of detection (LOD) for glucose of the proposed method was 0.003 mg/mL. The relative standard deviation (RSD) and the average recovery rate of the method was 0.04 and 100.3% , respectively, which denoted high repeatability and precision. Hence the established method is simple, economical, rapid and accurate, and suitable for routine analysis of bulk samples.
