The Effect of inorganic salts on the production of endo-β-D-1,4-glucanase from cellulose-degrading fungi

摘要: 本论文系统研究了不同种类无机盐对纤维素降解真菌产内切β-D-1,4-葡聚糖酶的影响,首先正交设计16个组合,具体为4因素(四种无机盐)2水平(两种浓度)并且考虑交互作用。以酶活为指标,通过进一步直观分析和方差分析,评价了各成分影响产酶的显著程度。结果表明,在本试验考察范围内,K2HPO4?3H2O对产酶有极显著正影响,而FeSO4?7H2O则相反;得到的优化组合为每升液体培养基中含0.1g CaCI2、0.1g FeSO4?7H2O、0.5g K2HPO4?3H2O和0.5g MgSO4?7H2O时,比未加无机盐培养基酶活提高111.916%;交互作用分析表示高浓度CaCI2、低浓度FeSO4?7H2O、高浓度K2HPO4?3H2O和高浓度MgSO4?7H2O 可最大化酶生产。

Abstract: The effect of inorganic salts on the production of endo-β-D-1,4-glucanase of cellulose-degrading fungi was investigated in this research. A orthogonal design was applied to design sixteen combinations which included four factors and two level, and the interaction was considered. Evaluating the impact significance of various components on the production of enzyme by the visual analysis and variance analysis with the index of enzymatic activity. The result showed that K2HPO4?3H2O had the positive remarkable effect on the production of enzme in the range of this research, but FeSO4?7H2O was the reverse. he optimized combination was as follows, 0.1g CaCI2, 0.1g FeSO4?7H2O, 0.5g K2HPO4?3H2O and 0.5g MgSO4?7H2O in every litre of liquid medium, the enzyme activity was improved 111.916% compared to the control without adding inorganic salts. The result of statistical analysis indicated that the low concentration of FeSO4?7H2O and the high concentration of K2HPO4?3H2O,CaCI2 and MgSO4?7H2O can achieve the maximum of enzyme production.
