阿魏蘑的DNA指纹分析与鉴定

摘要:

为及时制止阿魏蘑菌株混乱现象的产生,对国内现有的11株栽培常用的阿魏蘑菌株进行DNA指纹分析;以RAPD、ISSR、SRAP多态性分析的方法为主,用筛选出来的8条RAPD、ISSR、SRAP引物共扩增出116个较为清晰的DNA条带,然后进行聚类分析;结果表明,可将11个供试菌株分为四个群体:类Ⅰ:包括Pl. f 0007;类Ⅱ:包括Pl. f 0002;类Ⅲ:包括Pl. f 0003;类Ⅳ:包括余下的8个菌株。本研究为阿魏蘑的核心种质的构建和资源的科学保存提供了依据,并为规范阿魏蘑的生产奠定了基础。

Abstract:

To stop the strains confusion for the timely, the DNA fingerprint analysis was used in the study of 11 strains of Pleurotus ferula which popular culture in the country. We mainly used RAPD, ISSR, SRAP polymorphism analysis method, and the 8 primers of RAPD, ISSR, SRAP which We screened produced 116 band-types of DNA fragments, and cluster analysis lastly. The results showed that all strains could be clustered into four groups, ClassⅠ: Pl. f 0007; ClassⅡ: Pl. f 0002; ClassⅢ: Pl. f 0003; ClassⅣ: The remaining 8 strains. The study not only provides the evidence for the foundation of the core collections of Pleurotus ferula and the scientific preservation of resources, but also lays a foundation for regulating the production of Pleurotus ferula.
