山羊心脏型脂肪酸结合蛋白cDNA序列克隆及生物信息学分析

cDNA Clonging of Heart Fatty Acid Binding Protein Gene from Goat and its Bioinformatic Analysis

摘要:

本文通过RT-PCR技术克隆山羊心脏型脂肪酸结合蛋白的cDNA 全序列,并对cDNA序列的氨基酸同源性、理化性质、二级结构特征进行了预测和分析。结果显示,山羊H-FABP基因cDNA全长402 bp,推测的氨基酸序列与绵头同源性为100%;H-FABP蛋白属疏水性蛋白,呈为弱酸性;二级结构以β折叠为主;不含跨膜结构域,具有潜在的信号肽位点、多个其它信号位点以及细胞脂肪酸结合信号位点,从而为揭示H-FABP蛋白空间结构和生物学功能研究提供理论基础。

Abstract:

Full length cDNA sequence of heart fatty acid binding protein from goat was cloned by RT-PCR technology. The analysis and prediction on the amino similarity, physicochemical properties and secondary structure character were conducted from acquired sequences. The results showed that goat H-FABP gene cDNA was 402 bp in length and its homology rates of predicted amino acid sequence was 100% to sheep. H-FABP belong to hydrophilic and faintly acid protein. The secondary structure was mainly composed of βsheet. It had no transmembranous regions but possessed potential signal peptide, several other signal sites and cell fatty acid binding site. These results would provide theoretical base for revealing the protein space structure and biological function.
