高纯度牦牛子宫内膜基质细胞的分离培养及其鉴定

Isolation, culture and identification of high pure yak endometrium stromal cells

摘要:

为获得高纯度的牦牛子宫内膜基质细胞,对牦牛子宫内膜使用胶原酶消化,分离基质细胞进行体外培养,研究其生长特性,并对其进行免疫细胞化学染色鉴定。结果,牦牛子宫内膜在37℃条件下,以2 g/L的胶原酶Ⅱ消化2.5 h,更换新鲜消化液继续消化2.5 h,消化效果好;用74 μm滤网过滤,再经400 r/min离心和自然沉降去除上皮细胞,可分离得到大量的单个细胞,所得细胞贴壁后可见梭形和多角形2种形态,免疫细胞化学染色显示这2种形态的细胞均表达波形蛋白,阳性率可达95%以上,不表达角蛋白。结果证明已成功分离到高纯度的牦牛子宫内膜基质细胞。

Abstract:

To obtain high purity Yak Endometrium Stromal Cells, yak endometrium tissue was digested by collagenase, stromal cells was isolationed and cultured in vitro, the growth characteristics and immunocytochemical staining was studied. The results showed that the digest effect was good with 2 g/L collagenase Ⅱ at 37℃ working 2.5 h, following replacing with fresh 2 g/L collagenase Ⅱ working 2.5 h; epithelium cells were able to be wiped off through 74 μm filter, followed by 400 r/min centrifugalization and sedimentation, large numbers of single cells were obtained. Two shapes of cells with the former of spinder and polygonal were found when they became adherent. It was verified that this 2 shapes of cells were positive for vimentin, with 95% of positive rate, and negative for cytokeratin by immunocytochemical staining method. The conclusion can be drawn that purified yak stromal cells were obtained.
