超临界流体提取金银花中总黄酮研究初报

Pilot on Supercritical CO2 Extraction of Flavonoids from Lonicera japonica Thunb Leaves

摘要: 为探讨采用超临界CO2萃取技术萃取金银花中总黄酮的工艺条件;采用分光光度法测定总黄酮的含量,考察压力、温度、时间和夹带剂对总黄酮提取率的影响,并同热醇浸泡提取法、微波提取法和超声提取法的得率进行比较;得到超临界CO2萃取最佳条件为:压力30Mpa、温度40℃、时间120min和夹带剂用量4.5ml/g时,提取得率最高。同热醇浸泡提取法、微波提取法和超声提取法相比,其总黄酮得率分别为8.92%、9.56%、9.32%、和10.24%;超临界CO2提取方法同其他方法相比,得率高、时间短,是一种合适的提取金银花中总黄酮的方法。

Abstract: Effects of pressure, temperature, time and dosage of chemical preparation in supercritical CO2 technology on extract abilities of flavone from Lonicera japonica Thunb were Piloted in this paper. And the yield of the flavonids was determined by UV spectrophotometer . This paper also compared the method supercritical CO2 extraction with hot alcohol extraction, soaking microwave assisted extraction and ultrasonic assisted extraction .Results showed that following conditions were optimal: pressure 30.0MPa, temperature 40℃, time 120min and dosage of chemical preparation 4.5ml/g. Extraction yield of hot alcohol ,soaking microwave assisted extraction ,ultrasonic assisted extraction and supercritical CO2 technology extraction were 8.92%、9.56% 、9.32% and 10.24% separately, and supercritical CO2 technology was a good extraction method for its high yield and short times.
