几种槭属植物亲缘关系的ITS序列分析

摘要: 本文对槭属植物的8个种(含变种)核糖体 DNA 内转录间隔区(ITS)序列进行了PCR扩增和序列分析,扩增出约750bp左右的的序列(包含完整的ITS1、5.8S和ITS2序列及部分18S和26S rRNA基因片段。各样品的 ITS1长度为221~236 bp,ITS2 为 231~237 bp,含有多个特异性信息位点。并利用 ITS 序列为依据对8种槭属植物系统发育进行分析,从分子水平上阐明了8种槭属植物的亲缘关系,为挪威槭嫁接生产中适宜砧木的选择提供了理论依据。

Abstract: In this study, the sequences of nrDNA ITS from eight varieties of Acer species were analyzed by PCR amplification and sequencing. The sequences of PCR products were about 750 bp including the whole ITS 1, 5.8 s, ITS 2 sequence and partial 18 s, 26 s rRNA gene fragment. The length of ITS1 and ITS2 sequences were 221~236bp and 231~237 bp respectively. There were distinct variant sites between the ITS sequences. Phylogenetic analysis of eight varieties of Acer species was carried out on the basis of ITS and the homology relation among them was elucidated from the molecular level, which provide the basis for the selecting of rootstock in the grafting of Acer Platanoides ‘Royal Red’.
