古蜡梅的组织培养和试管苗的移栽及栽培技术

Tissue Culture of hundred year old - Chimonanthus praecox(L.)Link and Transplanting and Cultivating of Tube-plants

摘要: 以提高古蜡梅的组织培养和试管苗移栽及栽培效果为目的,用嘉定钱门塘的古蜡梅为起始材料,进行了无菌苗诱导、快速繁殖、生根和移栽试验。结果表明,1/2MS作为基本培养基明显优于Read、WPM、N6;诱导培养基中添加2.0mg/L 2iP、0.2mg/L IAA、0.5 mg/L GA3和1.5-5.0mg/L PVP可提高古蜡梅的繁殖系数和抑制外植体褐化;适当降低快繁培养基中的激素浓度可以防止玻璃苗的发生;添加0.5mg/L IBA和0.05mg/L NAA可使古蜡梅组培苗的生根率在75%左右。试管苗移栽前进行1-2天的炼苗和移栽后2周内保湿防晒可以提高成活率;抑制杂草可以促进试管苗健壮生长;控制地下水位可以提高试管苗的根系活力。

Abstract: The purpose of this research is to improve efficiency of tissue culture and transplanting and cultivating of tissue culture derived-plantlets of hundred year old-Chimonanthus praecox(L.)Link. Using annual branch as material, shoot inducing, rapid propagation, rooting and cultivation were studied. The results showed that the 1/2MS(1/2 of macroelements) as basic media was super to Read、WPM、N6, propagation coeifficient was increased and frequency of explants browning was decreased by supplemented with 2.0mg/L 2iP, 0.2mg/L IAA, 0.5 mg/L GA3 and 1.5-5.0mg/L PVP in inducing media, the number of vitreous buds were decreased by reducing hormone concentration, the frequency of rooting was 75% in media containing 0.5mg/L IBA and 0.05mg/L NAA. Transplanting survival rate of tube-plantlets could be improved by hardening plantlets for 1-2 days and maintained humidity for 14 days. The growth of tube-plants was promoted by holding weeds and root-vigor be increased by controlling the level of groundwater.
