猪流感H3N2亚型HA 基因真核表达载体构建及其对小鼠的免疫效果的研究

Immune efficacy of the recombination plasmids expressing HA gene of H3N2 subtybe SIV in mice

摘要:

采用PCR方法扩增出猪流感病毒四川分离株H3N2亚型HA全基因,将其克隆到真核表达载体pcDNA3.1(+)上,成功构建出真核重组质粒pcDNA3.1-HA。采用纯化后的pcDNA3.1-HA质粒和灭活全病毒以不同的组合免疫4-6周龄的Balb/c小鼠,免疫2次间隔3周,加强免疫3周后,小鼠接种致死量的H3N2病毒;检测免疫后小鼠的血清抗体水平及攻毒后肺部病毒残留。结果表明,pcDNA3.1-HA质粒和灭活全病毒免疫小鼠后,均能够产生一定的免疫效果,能为小鼠提供H3N2亚型猪流感病毒保护。

Abstract:

The HA gene of SIV amplified by PCR, was subcloned into eukaryotic expressing vectors pcDNA3.1.we examined the antibody response to DNA vaccine and inactivated virus and the protection against H3N2 SIV challenge using mouse model. Mice were immunized with different immune formation. Three weeks after the second immunization, mice were challenged with a high dose of H3N2 SIV. The ability of protection against virus challenge in mice was evaluated by induction of serum antibody response, the lung virus titers of mice. The results demonstrate the immune response induced by different immune formation in mouse model has been proved to be able to protect against the challenge with H3N2 SIV.
