桃‘豫农矮砧1号’快繁体系初代培养基的初探

Exploring of the Suitable Initial Culture in the Fast Reproduction System of ‘Dwarf Yunong Rootstock No.1’ of Prunus persica

摘要: 通过对不同完全培养基的筛选,进而探寻能够提高桃‘豫农矮砧1号’萌芽率的初代培养基,为桃‘豫农矮砧1号’快繁体系的建立打下坚实的基础,以桃‘豫农矮砧1号’的茎段为外植体,进行离体培养中初代培养基的筛选,以MS为基本培养基,附加不同浓度生长素IBA(3-吲哚丁酸)与细胞分裂素6-BA(6-苄基腺嘌呤)。结果表明:桃‘豫农矮砧1号’快繁体系中最佳初代培养基为MS+1.6 mg/L 6-BA+0.4 mg/L IBA,芽诱导率在IBA 0.1~1.0 mg/L,6-BA 0.8~1.6 mg/L的浓度范围内随激素浓度的上升而上升,并且以MS+1.6 mg/L 6-BA+0.4 mg/L IBA的芽诱导率最高。

Abstract: Explants of ‘dwarf Yunong rootstock No.1’ stem segments were cultured in vitro to select the most suitable initial culture. The MS was selected as the basic culture, plus different contents of IBA and 6-BA. By screening of different complete medium, and seeking the suitable initial culture which could improve germination rate of ‘dwarf Yunong rootstock No.1’, to establish the basis for peach ‘dwarf Yunong rootstock No.1’ rapid propogation system. The results showed that: the most suitable initial culture in the fast reproduction system was MS+1.6 mg/L 6-BA+0.4 mg/L IBA, and the bud induction was rising with the increase of the contents of hormone in the scope with IBA 0.1-1.0 mg/L, 6-BA 0.8-1.6 mg/L. In a word, the most suitable medium for the bud induction of stem segments was MS+1.6 mg/L 6-BA+ 0.4 mg/L IBA.
