5个绵羊品种BMPR-IB基因第7外显子多态性分析

Polymorphism of BMPR-IB Gene Exon7 in Five Populations

摘要: 为了探索绵羊多胎性的分子机理以及为绵羊繁殖力的标记辅助选择和育种提供理论依据。采用PCR-SSCP方法,对不同繁殖力的5个绵羊品种(特克赛尔羊、无角陶赛特羊、德国美利奴羊、小尾寒羊、藏羊)BMPR-IB基因第7外显子进行多态性检测。结果表明:5个绵羊群体均存在2种基因型,AA型和AB型;测序结果表明:AB型个体在该基因编码区第846位点发生C→T的突变,出现C/T的杂合。5个绵羊品种的优势基因型均为AA型,优势等位基因均为A基因。特克赛尔羊、无角陶赛特羊、德国美利奴羊多态信息含量均处于0.25~0.5之间,属于中度多态,小尾寒羊、藏羊属于低度多态(PIC<0.25);χ2适合性检验表明:除特克赛尔羊之外,其余4个绵羊品种均处于Hardy-weinberg平衡状态;显著性检验分析表明:该突变位点在不同繁殖力母羊群体中的分布有一定的差异。其中,特克赛尔羊、无角陶赛特羊以及德国美利奴羊3个品种的绵羊相互之间分布差异不显著(P>0.05),特克赛尔羊、无角陶赛特羊与小尾寒羊之间差异显著(0.010.05)。该突变位点可以作为控制绵羊繁殖力的潜在分子标记位点。

Abstract: For exploring the molecular mechanism of prolificacy and providing theoretical basis for marker-assisted selection and breeding of fecundity in sheep. PCR-SSCP was used to detect polymorphism of BMPR-IB gene Exon7 for five sheep breeds (Texel Sheep, Poll Dorset Sheep, Germany Merino Sheep, Small Tail Han Sheep, Tibetan Sheep) with different fertility. The results showed that AA and AB genotypes were found in five groups. The sequencing analysis indicated that AB genotype was C→T mutation in coding region of 846 and existed the heterozygous of C/T. Genotype AA was dominant genotype and allele A was dominant allele in five sheep breeds. The PIC of Texel Sheep, Poll Dorset Sheep and Germany Merino Sheep was between 0.25 to 0.5 and they were moderately polymorphic by population genetics, Small Tail Han Sheep and Tibetan Sheep were low polymorphism (PIC<0.25). In addition to the Texel Sheep, the other sheep breeds were in Hardy-weinberg equilibrium by χ2 test. This mutation point distributions had differences in different prolificacy sheep groups by significant test. Germany Merino Sheep had no significant differences with Texel Sheep and Poll Dorset Sheep (P>0.05), Small Tail Han Sheep had significant differences with Texel Sheep and Poll Dorset Sheep (0.010.05). This mutation could be considered as a potential locus to control reproduction of sheep.
