盐胁迫下杂交水稻种子发芽特性及耐盐性评价

Germination Characteristics of Hybrid Rice Seeds Under Salt Stress and Their Evaluation of Salt-Tolerance

摘要: 根据不同杂交水稻组合发芽期对盐胁迫的反应,筛选出适合在盐渍土壤种植的杂交水稻组合。采用人工气候箱内培养皿培养,用4、8和12 g/L 3个NaCl单盐浓度对8份杂交水稻组合进行盐胁迫处理,以蒸馏水处理为对照,每培养皿放30粒种子。分别于4天和7天后调查种子发芽率,7天后调查芽长、主根长和根数。以相对盐害率对杂交组合进行耐盐性评价。发芽率、芽长、主根长和根数随盐浓度升高总体呈下降趋势,但较低盐浓度(4、8 g/L)时的下降不明显,部分组合还高于对照;高浓度盐溶液(12 g/L)显著抑制水稻种子的发芽率,芽长、主根长和根数均显著下降,降幅分别为35.8%、57.6%、76.1%和83.8%;随着盐浓度的提高,根数下降较快,盐胁迫对杂交水稻组合的根数影响要大于芽长和根长;筛选出发芽期强耐盐组合‘桂源优500’、‘II优1040’(1级),‘II优1050’、‘甬优12号’(3级)。以12 g/L NaCl单盐浓度可作为杂交水稻组合发芽期耐盐性分级评价的适宜浓度,相对盐害率可以作为杂交水稻发芽期耐盐性的评价指标。

Abstract: The aim of the study were to explore response of different hybrid rice seeds to salt stress, screen hybrid rice combinations suitable for growing in saline soils. Thirty seeds of each of 8 hybrid rice combinations were treated with 0, 4, 8 and 12 g/L NaCl and incubated in Petri dishes for 7 days. Germination rate of hybrid seeds were counted in 4 and 7 days after germination, and bud length, main-root length, number of roots were investigated after 7 days. Salt tolerance (ST) of these combinations was evaluated by the relative damage rate. The results showed that germination rate, bud length, main-root length, number of roots were decreased along with increase of salt concentration.The germination rate was obviously inhibited by high concentration of salt solution (12 g/L), and the germination rate, bud length, main-root length, number of roots were decreased by 35.8%, 57.6%, 76.1% and 83.8%, respectively. However, the four characters didn’t show obvious decrease under lower salt concentration (4, 8 g/L), and inversely they showed increase in some combinations. Salt stress had greater influence on number of roots than bud length and main- root length, in which number of roots decreased more rapidly along with rising of salt concentration.‘Gui-yuan-you 500’,‘II-you 1040’,‘II-you 1050’and‘Yong-you 12’were screened out as salt tolerance hybrid combinations with tolerant score of 1, 1, 3 and 3, respectively. The concentration of salt solution (12 g/L) was suitable to screen ST for hybrid seeds at germination stage, and the relative damage rate was recommended as an appropriate index for evaluation of ST.
