上海地区虾源弧菌分离鉴定及分子分型研究

Identification and Molecular Typing of Vibrio Strains Isolated from Litopenaeus vannamei in Shanghai

摘要: 为了进一步了解上海市凡纳滨对虾主养区虾源弧菌的分布特征以及主要种属的分子基因型特征,为本地区虾的弧菌病的流行调查、防控治疗提供依据。本研究采用选择性培养基筛选和全自动细菌鉴定仪鉴定的方法,对不同时间,不同池塘的虾源弧菌进行分离鉴定,总共分离到52株不同来源的弧菌,其中30株副溶血弧菌(57.7%),9株拟态弧菌(17.3%),7株创伤弧菌(13.5%),4株溶藻弧菌(7.7%),1株霍乱弧菌(1.9%)以及1株河流弧菌(1.9%)。调查结果显示,上海地区虾源弧菌以副溶血弧菌为主。进而对30副溶血弧菌株副溶血弧菌进行分子分型研究,EIRC-PCR指纹图谱显示基因型存在多态性,每个分离株的基因扩增条带数介于4~8之间,大小都在250~4500 bp之间;聚类分析分为5个亚群,其中Ⅰ亚群15株(50%),Ⅱ亚群6株(20%),Ⅲ亚群6株(20%),Ⅳ亚群2株(6.7%),Ⅴ亚群(3.3%)。结果表明,上海地区虾源副溶血弧菌存在一种主要流行亚群,对生产中药物筛选及疫苗研制具有一定的参考价值。

Abstract: The study aims to understand theVibrio distribution characteristics ofLitopenaeus vannamei and the molecular genetic characteristics of main species in Shanghai, and provide a basis for the investigation and prevention of vibriosis in the future. The isolation and identification ofVibrio fromLitopenaeus vannamei was studied by selective medium separation and automatic identification system of bacteria. 52 strains were isolated from different fishponds and different regions. Among these strains, 30 strains were Vibrio parahaemolyticus (57.7%), 9 strains wereVibrio mimicus (17.3%), 7 strains wereVibrio vulnificus (13.5%), 4 strains wereVibrio alginolyticus (7.7%), 1 strain wasVibrio cholerae (1.9%) and 1 strain wasVibrio fluvialis (1.9%). The results of the investigation showed thatVibrio parahaemolyticus was the main type ofvibrios which was isolated fromLitopenaeus vannamei in Shanghai. The genetic typing of 30Vibrio parahaemolyticus strains carried out through ERIC, ERIC-PCR finger print revealed that the genotypes had abundant genetic diversity. And every strain was amplified 4-8 bands within 250-4500 bp. 5 cluster groups were divided by cluster analysis, including 15 strains of Ⅰcluster group(50%), 6 strains of Ⅱ cluster group (20%), 2 strains of Ⅲ cluster group (6.7%), 2 strains of Ⅳ cluster group (6.7%), and 1 strain ofⅤ cluster group (3.3%). The results indicated thatV. parahamolyticus of Ⅰcluster group was popular in Shanghai.
