适合于甜菜品种鉴定的ISSR核心引物的筛选

Screening of ISSR Core Primer Suitable for Identifying Beet Cultivar

摘要: 探寻适合甜菜品种鉴定的ISSR引物,用以构建甜菜品种的指纹图谱。利用一个品种对全部100条ISSR引物进行退火梯度实验,然后利用16个进口甜菜品种在每个引物的最佳退火温度下对全部引物进行扩增,筛选适合甜菜品种纯度鉴定的ISSR引物。结果从100条ISSR引物中筛选出多态性高、扩增条带清晰、重复性好的引物16条,这16条ISSR引物总共扩增100条多态性条带,引物的PIC值均大于0.6,从理论上讲,这些核心引物完全可以实现对现有甜菜品种的鉴定。

Abstract: The aim was to seek appropriate ISSR primers of beet identification and then build beet fingerprint. The author utilized one species to do annealing gradient experiment on 100 ISSR primers and then expanded all primers under their best annealing temperature using 16 imported beet species, appropriate beet species was selected for purity identification. As a result, 16 primers with high level of polymorphism, clear amplification bands and good repetitiveness were selected which could expand to over 100 polymorphic bands in total with PIC over 0.6 on average. Theoretically speaking, these core primers could accomplish the identification of beet species.
