不同品种羊miR-1298-5p及其潜在靶基因TGF-βR1表达的比较研究

Comparative Study on Expression of miR-1298-5p and its Potential Target Gene TGF-βR1 in Fine-wool Sheep and Cashmere Goats

摘要: 旨在探讨miR-1298-5p 与TGF-βR1 在不同品种羊皮肤毛囊不同发育时期的潜在关系,为研究miR-1298-5p 与TGF-βR1 对皮肤毛囊发育作用提供理论研究数据。以绒山羊和细毛羊皮肤毛囊组织为材料,通过生物信息学方法预测miR-1298-5p 的靶基因,采用qRT-PCR对miR-1298-5p 与其潜在靶基因TGF-βR1 进行核酸水平表达检测,利用Western blot 对潜在靶基因TGF-βR1 进行蛋白水平表达检测。生物信息学预测结果表明TGF-βR1 的3''UTR 存在miR-1298-5p 种子区结合位点。miR-1298-5p 在绒山羊和细毛羊皮肤毛囊发育的生长期到退行期相对表达量上调而退行期到休止期相对表达量下调,miR-1298-5p 在不同品种羊皮肤毛囊不同发育时期呈现差异性表达。绒山羊皮肤毛囊发育生长期TGF-βR1基因mRNA的相对表达量高于细毛羊,而退行期和休止期低于细毛羊;从表达趋势上看,TGF-βR1 与miR-1298-5p 表达趋势相反,呈现负调控趋势,说明TGF-βR1 可能为miR-1298-5p 的靶基因。TGF-βR1蛋白在绒山羊皮肤毛囊发育生长期的相对表达量高于细毛羊,而退行期和休止期则低于细毛羊,与核酸水平相对表达量趋势相同,但都与miR-1298-5p 表达趋势相反,更进一步说明TGF-βR1 可能为miR-1298-5p的靶基因。

Abstract: The study aimed to investigate the regulation of miR-1298-5p and its potential target gene TGF-βR1 on skin and hair follicles development at different stages in fine-wool sheep and cashmere goats for obtaining theoretical research data on miR-1298-5p and TGF-βR1 in the development of skin and hair follicles. Using skin and hair follicles tissues from cashmere goats and fine-wool sheep at different stages as materials, potential target genes of miR-1298-5p were predicted by bioinformatics analysis. Expression of miR-1298-5p and its potential target gene TGF-βR1 were determined by qRT-PCR. Expression of TGF-βR1 protein was determined by Weston blot. Bioinformatics analysis showed that 3''UTR of TGF-βR1 from fine-wool sheep and cashmere goats presented a binding site of miR-1298-5p. Relative expression of miR-1298-5p from fine-wool sheep and cashmere goats were both up regulated from anagen to catagen and down regulated from catagen to telogen, and presented difference between fine-wool sheep and cashmere goats. Relative expression of TGF-βR1 mRNA in cashmere goats was higher than that in fine-wool sheep at anagen and lower at catagen and telogen. And miR-1298-5p and TGF-βR1 showed a negative regulation trend, which implied that TGF-βR1 might be the target gene of miR-1298-5p. Relative expression of TGF-βR1 protein in cashmere goats was also higher than that in fine-wool sheep at anagen and lower at catagen and telogen, being the same as the relative expression of TGF-βR1 mRNA, which further implied that TGF-βR1 was target gene of miR-1298-5p.
