哈蟆油蛋白多肽的透皮作用研究

摘要:

比较酶解前后哈蟆油蛋白多肽的相对分子质量、黏度和透皮情况。采用胃蛋白酶-碱性蛋白酶双酶法制备哈蟆油蛋白多肽,应用旋转粘度计测定酶解过程中黏度变化;以离体小鼠的皮肤为透皮屏障,应用透皮吸收仪测定不同透皮时间透皮液中蛋白多肽的透皮率和分子量分布。双酶法制备的哈蟆蛋白多肽黏度从5278.85 mPa·s(1.0 g/100 mL)降到12.56 mPa·s(7.0 g/100 mL),分子质量1147 kDa蛋白质从75.36%降到8.83%,透皮率从4.50%增加到9.14%。透皮36 h时,透皮液中主要为分子质量小于1 kDa哈蟆油蛋白多肽约占62%,其次是分子质量2.46 kDa的蛋白多肽占13.34%。双酶法制备的哈蟆油蛋白多肽分子量低、水溶性好,易于透过皮肤吸收。

Abstract:

A comparative study on relative molecular mass, viscosity and skin permeation effect of ranae oviductus peptides before and after enzymolysis was carried out. Ranae oviductus peptides were prepared through hydrolyzing ranae oviductus protein by pepsin and alkaline protease. The viscosimeter was used to detect the changes of viscosity during the enzymatic hydrolysis process. Taking the mouse skin as a transdermal barrier, percutaneous rate and the relative molecular weight distribution of percutaneous peptides at different times were detected by apparatus of transdermal absorption and high performance liquid chromatograph (HPLC), respectively. The results showed that the viscosity of ranae oviductus peptides decreased from 5278.85 mPa·s (1.0 g/100 mL) to 12.56 mPa·s (7.0 g/100 mL); the peak area percentage of ranae oviductus proteins with the relative molecular mass 1147 kDa went down from 75.36% to 8.83%, however, the percutaneous rate of ranae oviductus peptides increased from 4.50% to 9.14%. When the percutaneous time was 36 h, percutaneous peptides with the relative molecular mass less than 1 kDa were the main component in percutaneous fluid, accounting for about 62%, followed by percutaneous peptides with the relative molecular mass of 2.46 kDa, accounting for 13.34%. In general, the ranae oviductus peptides prepared by double-enzyme method have low molecular weight and good water solubility, and could penetrate the skin of mouse easily.
