非洲紫罗兰离体快繁体系的建立

Study on Tissue Culture Technique of Saintpantia ionantha

摘要:

本研究以非洲紫罗兰的带腋芽茎段为外植体,研究非洲紫罗兰组织培养的最佳培养基组合,诱导侧芽生长的最佳激素浓度配比,以及诱导侧芽生根的最佳激素及浓度。实验以CC、N6、MS、B5等作为基本培养基,将其无机大量、有机物、无机微量作为3个不同的影响因子,添加不同浓度的6-BA和NAA进行侧芽诱导;再以不同浓度的NAA、IAA、IBA组合,诱导侧芽生根。实验结果表明:非洲紫罗兰生长的最佳培养基组合为MS大量+CC微量+B5有机,侧芽增殖的最佳培养基为MS大量+CC微量+B5有机+0.5~1.0 mg/L 6-BA+0~0.1 mg/L NAA,诱导侧芽生根的最佳培养基为1/2MS+0.1 mg/L NAA或1/2MS+0.1 mg/L IAA。

Abstract:

The optimal combination in tissue culture of Saintpantia ionantha was discussed in this study. Stem and axillaries of S. ionantha were used as explant for tissue culture, the basic culture medium included CC, N6, MS and B5, and major elements, organics and trace elements were selected as three different impact factors, the different concentrations of 6-BA and NAA were used to induce lateral bud, then different concentrations of combinations of NAA、IAA and IBA were used to induce root. The results show that: the optimal culture medium combination for S. ionantha is MS major elements + CC trace elements + B5 organics; the optimal combination and the concentrations for proliferating lateral bud is MS major elements + CC trace elements + B5 organics+0.5-1.0 mg/L 6-BA + 0-0.1 mg/L NAA; and the optimal culture medium combination and the concentrations for inducing root is 1/2MS+0.1mg/L NAA or 1/2MS+0.1 mg/L IAA.
