40个龙眼品种(品系)DNA指纹图谱构建及遗传关系分析

DNA Fingerprinting of 40 Dimocarpus longan Germplasms by ISSR Markers and Analysis of Genetic Relationship

摘要: 有效鉴别龙眼新品种(品系),为龙眼种质资源研究和新品种保护提供理论基础。利用ISSR分子标记分析‘冬宝9 号’、‘高宝’等4 个龙眼新品种(品系)与36 个龙眼品种的遗传关系,并建立其DNA指纹图谱。从80 个ISSR 引物中筛选出11 个多态性好、扩增清晰的引物。结果显示,11 个引物共获得146个位点,有125 个多态性位点,多态性百分率为85.7%。40 个龙眼品种(品系)的遗传相似系数在0.58~0.88 之间。UPGMA聚类结果表明,40 个龙眼品种(品系)在遗传相似系数为0.64 的水平处划分为3 组。引物UBC810/UBC818 组合可将40 个龙眼品种(品系)全部区分开,并依此建立了数字化的DNA指纹图谱,为龙眼新品种的鉴别、分类、示范及推广等方面提供依据。

Abstract: To differentiate and identify varieties and types of Dimocarpus longan germplasm, the inter-simple sequence repeat (ISSR) method was applied to assess the genetic variations of 40 Dimocarpus longan varieties including 4 new varieties. From 80 ISSR primers, 11 were employed on 40 samples. Cluster analysis revealed that the genetic similarity coefficient ranged from 0.58 to 0.88, and the 40 varieties could be clustered into 3 groups using a genetic similarity coefficient threshold of 0.64. The 40 varieties could be distinguished completely by 2 ISSR primers (UBC810/UBC818), and digital DNA fingerprint was established accordingly. This study provided an important basis for the identification of new varieties, and the evaluation and utilization of germplasm resources.
