甘蓝枯萎病抗性鉴定方法研究

Study on the method of Fusarium wilts resistance in cabbage

摘要: 甘蓝枯萎病是由镰刀菌(Fusarium oxysporum f. sp. Conglutinans)引起的土传病害,属于我国新发病害,近年来已发展成为中国北部夏秋甘蓝生产区的毁灭性病害,并且发病地域逐年扩大。抗病品种的使用是目前减少甘蓝枯萎病损失的最有效方法。而探索高效的甘蓝抗枯萎病鉴定方法以准确鉴定甘蓝种质资源的相应抗性基因是抗病育种的基础性工作。因此,我们在前期病原物分离和鉴定工作的基础上,采用来源于山西寿阳的强致病力株GLHW1作为病原,通过因子设计方法系统分析了3个接种方法,3个接种浓度及2个接种时期的接种效率及不同因素间的互作效应。研究结果表明:3个因素对接种效果的重要性影响依次为接种方法>接种浓度>接种时期,推荐给甘蓝育种者的接种程序是:在3叶1心期,将幼苗的根系浸入每毫升含1×106个孢子的悬浮液中15分钟。

Abstract: Fusarium wilts, caused by Fusarium oxysporum f. sp. conglutinans, is becoming a destructive disease of warm season-grown cabbage in North of China.The aim of this paper was to explore an optimized method for screening and identifying disease resistant and susceptible cabbage genotypes. In this study, we took the most virulent isolate from Shanxi, named GLHW1, as the inoculum for optimized method screen. A factorial experiment was conducted to evaluate the effectiveness of different screen method. Based on the results of present study, the inoculation procedure recommended to cabbage breeders is dipping root of cabbage seedling at the 3-true-leaf stage with a suspension of spores containing 1×106 spores per milliliter.
