Molecular Detection and Analysis of 1Dx5+1Dy10 in Partial Wheat Cultivars(Lines) From Huang-Huai Wheat Region of China

摘要:

目前在已报道的检测高分子量麦谷蛋白亚基1Dx5的PCR分子标记中,由引物Dx-F,Dx5-F和Dx-R组成的共显性标记,能稳定地扩增出5亚基和非5亚基的特异序列,并且无非特异PCR产物,是用于高分子量麦谷蛋白亚基1Dx5分子检测的最理想标记。本次实验利用此对引物, 通过PCR技术对221份小麦亲本进行了1Dx5亚基检测。结果表明,在所检测的材料中有35份材料携带1Dx5亚基,占所测材料总数的16.3%,远低于国外的基因频率(约85%)。

Abstract:

At present, reported at detection of HMW-GS 1Dx5 molecular markers, the marker,composed of three primers Dx-F, Dx5-F and Dx-R, is a specific co-dominant PCR markers for 1Dx5 subunit,and has no non-specific amplicon.It was strongly recommended to be used in wheat molecular assiatant selection (MAS) breeding for good bread-marking quality. The experimental use of this pair of primers by PCR technology for 221copies of a parental wheat gene 1Dx5 subunit. The results showed that the detection of 35 copies of Materials Materials 1Dx5subunit carried, representing the total number of measured Materials16.3%, much lower than the frequency of foreign genes (about 85%).
