一株产胶原酶海洋微生物发酵条件的研究

Fermentation Conditions of a Collagenase-Producing Marine Microorganism

摘要:

于海洋污泥中筛选得到的Aeromonas sp.F3所产的胞外酶对胶原蛋白有水解作用,采用单因素试验法对Aeromonas sp.F3的发酵条件及培养基组分进行全面优化,提高该胶原酶生产菌的产酶效率。经优化试验结果表明,该菌株最佳发酵条件为:培养时间24 h,温度40℃,初始pH为8,装瓶量80~100 mL/250 mL;培养基组分为:葡萄糖3 g/L,酵母浸粉9 g/L,NaCl 5 g/L,MnSO4 10 mg/L。经优化试验明显提升了该菌株的产酶能力。

Abstract:

Aeromonas sp.F3 screened from marine mud should secrete a protease to hydrolyze collagen. The fermentation of Aeromonas sp.F3 was investigated through single-factor test. The main factors included fermentation conditions and the medium. With single factor trial, the optimal culture condition was determined: 40℃ of fermentation temperature, pH8 and volume 80~100 mL/250 mL for 24 h. The optimal liquid medium was: 3 g/L glucose as carbon source, 9 g/L yeast extract powder as nitrogen source, 5 g/L NaCl as inorganic salt and 10 mg/L MnSO4 as metal ion. Under the optimum condition, the enzyme activity increased very observably.
