二次正交旋转组合设计优化雷公藤继代增殖培养基

Optimize the multiplication medium of Tripteryguium wilfordii Hook.f. through the quadratic orthogonal rotation combination design

摘要: 摘 要:为获得雷公藤试管苗高频继带增殖的最佳条件,采用二次正交旋转组合设计对其继代增殖培养基进行优化研究。结果表明:雷公藤继带苗增殖倍数(y)对MS浓度(x1)、6-BA浓度(x2)及NAA浓度(x3)3个试验因子的正交回归模型为y=6.98483-0.25481x12-0.51998x22-0.37856x32-0.35x1x2-0.325x2x3。对模型的进一步解析表明,3个因素在试验设定范围内对增殖倍数的影响存在低浓度促进高浓度抑制的现象。MS在0.398~0.602倍常规浓度下,6-BA和NAA的浓度分别在0.919~1.092 mg/L和0.167~0.233 mg/L时,其增殖倍数大于6.3的可靠性为95%。通过实际接种验证,30 d平均增殖倍数为6.53,证明本研究优化出的培养基是雷公藤试管苗增殖的最佳培养基。

Abstract: Abstract: The aim of the study was to get the best condition of multiplication culture of Tripterygiium Wilfordii Hook.f., the multiplication medium of Tripterygiium Wilfordii Hook.f. was optimized with quadratic orthogonal combination design. The results showed that the quadratic orthogonal recursive model of multiplication rate(y) to three concentration factors of MS(x1), 6-BA(x2) and NAA(x3) is y=6.98483-0.25481x12-0.51998x22-0.37856x32-0.35x1x2-0.325x2x3. The results showed that the low concentration of MS, 6-BA and NAA promoted the rate of multiplication and the high concentration inhibited it; and that the rate of multiplication was more than 6.3 with the probability of 95% when the MS concentration was in range of 0.398 to 0.602 MS, 6-BA in 0.919 to 1.092 mg/L and NAA in 0.167 to 0.233 mg/L.
