摘要: 古代DNA的研究方兴未艾,本文概述了古DNA技术在考古学研究中一些应用,并且利用古DNA技术初步研究了“南澳Ⅰ号”古沉船出水的古代腐肉样品。用通用引物18SrDNA及动物组织线粒体(mtDNA 16S rRNA)采用多种条件进行扩增分析,根据实验结果对水古代腐肉DNA的研究方法及结果进行探讨,为以后更多的出水遗存材料的古DNA研究提供借鉴。

Abstract: Since many studies have highlighted the feasibility of analyzing DNA from all type of ancient remains including bones, hair, teeth and even coprolithes. Therefore aDNA(ancient DNA) is widely investigated in archeaozoology research fields. However the study of aDNA is challenging due to the contamination by exogenous current DNA. In this paper, we tried to extract the ancient DNA from the ancient meat of Nan’ao Ⅰ shipwreck, and amplify the aDNA by Universal Primers of 18SrDNA and mtDNA 16S rRNA (animal tissue). The designated primers successfully amplified the aDNA .But the results were not reasonable . Through the analysis of the effects of experimental factors, to provide reference for the further research.
