基于变温层积的激素处理对大百合种胚后熟的影响

Influence of Hormone Treatment on Cardiocrinum giganteum Seed After-ripening During Variable Temperature Stratification

摘要: 为解决大百合种子胚后熟引起的休眠问题,以云南哀牢山自然保护区大百合种子为试验材料,采用不同激素结合变温层积处理的方法对大百合种子进行研究。结果表明,在大百合种子变温层积90天后加入6-BA 50 mg/L浸泡24 h 能促进胚生长,胚率为45.873%(层积120 天后)、萌发率45.33%(从第1粒种子萌发开始30天后统计萌发率),发芽时间较空白对照提前6天,同样层积条件下加入6-BA 20 mg/L的胚率为23.008%,萌发率为30.33%,加入SNP 13.9 mg/L 的胚率为27.163%,萌发率为31.33%;空白对照胚率为16.741%,萌发率为26.67%。综合胚率、萌发率和发芽时间筛选出层积90 天后加入6-BA50 mg/L浸泡24 h有利于大百合种子胚后熟并促进种子萌发。

Abstract: The paper aims to solve the problem of dormancy of Cardiocrinum giganteum seed which is induced by the embryo afterripening. The study was carried out by means of different hormone and temperature stratification using the seed of Cardiocrinum giganteum from the Ailao Mountain Nature Reserve in Yunnan. The result showed that the addition of 50mg/L of 6-BA for soaking of 24h can promote the growth of embryo after 90d of temperature-accelerated stratification of seed of Cardiocrinum giganteum, the embryo rate was 45.873% after 120d of stratification, and the germination rate was 45.33% which was counted after 30d of germination of the first seed, and the germinating time reduced 6 days than blank control group,with the addition of 20mg/L of 6-BA, the embryo rate was 23.008% and the germination rate was 30.33% after 90d of temperature-accelerated stratification,with the addition of 13.9mg/L of SNP, the embryo rate was 27.163% and the germination rate was 31.33%.,the embryo rate of blank control group is 16.741% and the germination rate was 26.67%. In view of the embryo rate, germination rate and germinating time, the addition of 50mg/L of 6-BA after 90d of stratification for soaking of 24h would be beneficial to promoting embryo after-ripening of seed of Cardiocrinum giganteum and to hastening seed germination.
