β-甘露聚糖酶高产菌株的紫外线诱变选育

Screening of β-mannanase High Yield Strain by Ultraviolet Mutation

摘要: 为给生产实践提供具有更高产酶量的优良菌株,本研究以实验室保藏的一株地衣芽孢杆菌HDYM-04为原始出发株,经自然筛选、紫外线诱变,选育出一株β-甘露聚糖酶高产菌株U2-12。结果表明:依据致死率所选用的最佳诱变剂量为照射距离50 cm,照射时间为10 s,此时,菌株的总突变率为76.11%,正突变率为54.60%,负突变率为20.92%,地衣芽孢杆菌U2-12的产β-甘露聚糖酶能力是原始未紫外线诱变的出发株的242.19%,其遗传性状稳定。本研究可以为后续试验及生产实践提供优势产酶菌株,并为研究紫外线对β-甘露聚糖酶产酶基因的影响提供先决条件。

Abstract: In order to provide strains which have a high enzyme yield, the original strain Bacillus licheniformis HDYM- 04 was isolated naturally and then treated with ultraviolet ray. The best mutation condition was determined to expose the strain to the ultraviolet source at 50 cm for 10 s based on the fatality rate. The total strain mutation rate was 76.11%, the positive mutation rate was 54.60%, and the negative mutation rate was 20.92%. A stable β-mannanase producing strain Bacillus licheniformis U2-12 was obtained with the enzyme activity being 242.19% of the original strain. The results can provide superior enzyme production strains for the following test and production practice, and references for studying the effect of ultraviolet ray on β-mannanase gene.
