甜菜夜蛾非典型嗅觉受体基因OR2的组织特异性和时空表达

Tissue-Specific Expression and Temporal and Spatial Expression of Atypical Odorant Receptor Gene OR2 in Spodoptera exigua (Hübner)

摘要:

本研究采用反转录多聚酶链式反应(RT-PCR)和实时定量PCR(Real-time quantitative PCR)技术对甜菜夜蛾Spodoptera exigua (Hübner)成虫不同羽化时期,不同部位组织内的非典型嗅觉受体OR2(SexiOR2)的组织表达谱和时空表达量进行了研究。结果表明,SexiOR2在甜菜夜蛾成虫的雌、雄蛾触角和喙内均表达。羽化后36 h的雄虫触角中的SexiOR2表达量最大,约为同期雌虫的5.5倍。SexiOR2在雌、雄虫喙内表达量极低,其他组织中均未发现表达。本研究明确了甜菜夜蛾非典型嗅觉受体OR2在不同性别、羽化时期和组织的表达情况。

Abstract:

To study the tissue-specific expression and quantitative analysis of atypical odorant receptor gene OR2 (orthologue to the Drosophila melanogaster DOR83b) in Spodoptera exigua (Hübner) (SexiOR2), the researchs on mRNA expression level and specific of atypical odorant receptor OR2 at different developmental stages in S. exigua were performed by reverse transcription polymerase chain reaction (RT-PCR) and real-time quantitative polymerase chain reaction (Real-time quantitative PCR). The results indicated that SexiOR2 was expressed in antennae and proboscis of male and female S. exigua. The expression level was the highest in male antennae 36 h after emergence, and was 5.5-fold higher than female antennae. The level of SexiOR2 was very low in proboscis. In this study, the tissue expression pattern and level of SexiOR2 was inferred at different developmental stages.
