三黄鸡β-防御素基因片段的克隆与序列分析

摘要:

根据GeneBank公布的鸡β-防御素序列设计一对特异性引物,运用RT-PCR技术从新郑三黄鸡肺脏组织中扩增其β-防御素基因并测序分析,结果表明,获得了195bp的cDNA目的片段,编码65个氨基酸;BLAST分析表明,该片段与GeneBank中已公布的鸡β-防御素Gal-9序列基因同源性达到98%。其与已公布的10个Gal-9序列进行同源性比较,并绘制系统发育进化树的结果显示,所测的防御素基因核苷酸同源性为91.8%~94.4%;进化树分析表明获得的目的基因与Gal-9基因处于同一进化分支。这为进一步研究Gal-9基因在地方品种鸡中的先天性免疫功能奠定了基础,也为研发具有广谱抗菌活性的新型肽类生物制品奠定了前期基础。

Abstract:

According to chicken β-defensin sequences published in GeneBank, a pair of specific primers was designed. The Gal-9 gene was amplified by using RT-PCR from the lung tissue of Xinzheng Sanhuang chicken. Then it was sequenced. The results showed that the length of the cDNA fragment obtained was 195bp, encoding 65 amino acids; BLAST analysis showed that the homology between the fragment cloned and the chicken β-defensin-Gal-9 gene sequences published in the Gene Bank was 98%. Then the sequence was contrasted with the 10 Gal-9 sequences published, at the same time, the phylogenetic tree was drawn. The results showed that the homology of them is from 91.8% to 94.4%; Phylogenetic tree analysis showed that the gene and Gal-9 gene in the same evolutionary branch. That not only provided a base for research on immunologic function of Gal-9 in the native chicken, but also laid the foundation of developing new antimcrobial product with broad spectrum.
