4个梅花品种的胚培养及愈伤组织诱导研究

Research on Embryo Culture and Callus Culture of Prunus mume Cultivars

摘要:

为保存和扩繁梅花种质资源,试验筛选4个梅花品种,取胚进行胚培养,旨在建立梅花胚培养的无菌体系。以梅花(Prunus mume Sieb. Et Zucc.)天然杂交的未成熟胚为外植体,初步探索‘淡寒红’、‘南京红须’、‘双碧垂枝’、‘粉瓣’等梅花品种的胚培养体系,并在此基础上,利用叶片建立了高效的愈伤组织诱导体系。结果表明:胚培养的最适培养基为MS+6-BA 1.0 mg/L+IBA 0.2 mg/L,叶片愈伤组织诱导培养基为MS+2,4-D 1.0 mg/L,最适的叶龄为幼叶时期。梅花的胚培养中,不同品种的萌发率具有一定差异,难易顺序是单瓣品种群<垂枝品种群<朱砂品种群。

Abstract:

In order to conserve and multiply germplasm rescources of Mei flower, the gern-free system of embryo-culture was constructed in seeds of four cultivars. The contents of this paper were as follows: we conducted immature embryos culture of Prunus mume ‘Danhanhong’, ‘Nanjing Hongxu’, ‘Shuangbi Chuizhi’, ‘Fenban’ derived from pollination. In addition, using leaf of seeding cultivation as explants, the efficient of callus culture was conducted. Results showed that the perfect culture medium was MS+6-BA 1.0 mg/L+IBA 0.2 mg/L. The optimum callus culture was MS+2,4-D 1.0 mg/L and perfect leafage was young period. Germination Rate of cultivars is different. The difficulty was single-lobe Mei Branch < hanger wood Mei Branch < cinnabar Mei Branch.
