Optimization of SDS-PAGE and Western-blot for Total Protein in Seedling Young Leaves of Lactuca sative L.

摘要:

为建立一套适合叶用莴苣(Lactuca sative L.)热激蛋白研究的SDS-PAGE和Western-blot方法。以叶用莴苣W7为材料,采用改进的TCA/Acetone法提取叶片蛋白,比较了两种电压下SDS凝胶分离蛋白质的效果,使用4℃保存两周的抗体稀释液并且得到了较清晰的热激蛋白70和内参蛋白Rubisco条带,初步确定了叶用莴苣叶片热激蛋白的western-blot实验条件。

Abstract:

In order to establish a method SDS-PAGE and Western-blot suitable for heat shock protein study of Lactuca sative L., with leaves of Lactuca sative L. (W7) as material, we used improved TCA/Acetone method extracted leaf protein, compared protein separating effect in SDS gel in two different volt, obtained clear bands of HSP70 and rubisco using anti-body diluent stored 2 weeks at 4℃, optimized and preliminary established Western-blot experiment condition of heat shock protein of Lactuca sative L. young leaves.
