牛心朴子总生物碱提取及精制方法研究初报

摘要: 以总生物碱含量为指标,考察提取溶剂浓度(以盐酸、乙醇为溶剂时)和提取方法(以水为溶剂时)对牛心朴子总生物碱提取量的影响;同时以吸光度为指标,研究了絮凝剂种类、投入量、絮凝体系温度、絮凝体系pH值对粗提液进一步精制的影响。结果表明,用60%乙醇回流提取时,其总碱含量最高;用1号澄清剂在50℃、pH=4~5、加入粗提液的20%时,澄清效果最好。这可初步作为牛心朴子生物碱提取及精制的最佳条件。

Abstract: Using total alkaloids as the guide, influencing factors of extraction yields of alkaloids from Cynanchum komarovii including solvent concentration (using hydrochloric acid and ethanol as extract solvent) and extracting method (using water as extract solvent) were studied. And using light absorbing range as the guide, the influencing factors of purification of the crude extract such as kind of flocculant, dosage of flocculant, temperature of flocculating system, pH value of flocculating system were studied based on single factor test. The results showed that the obtainment of total alkaloids were highest when extract with 60% ethanol, and the optimal flocculating conditions were: under 50℃, pH value were 4~5, adding the NO.1 flocculant as 20%(v/v) of crude extract.
