Preliminary Studies on Tissue Culture of Pistachio

摘要: 阿月浑子是中国西部与华北地区极有发展前景的坚果树种,但其微繁技术至今未有突破,以7年生阿月浑子嫁接树茎段为外植体,进行启动培养试验,对MS、改良MS、Q-L4、DKW 4类培养基,BA、IBA、NAA、GA 4种激素与用量,7%次氯酸钠和0.1%升汞溶液与灭菌时间、葡萄糖和蔗糖与用量、叶酸、D-泛酸钙与LH(水解乳蛋白)3种添加剂与用量的培养效果进行比较分析,确定阿月浑子外植体0.1%升汞溶液灭菌10min 效果最好,最优培养基为1/2DKW+0.5mg/l 6-BA +0.5mg/l IBA + mg/l GA3 +1mg/l叶酸+ 300mg/l LH +1mg/l D-泛酸钙+15g/l葡萄糖+ 6.5g/l琼脂。

Abstract: Pistachio is a potential nut tree in the north and west of China, however, the micropropagation of pistachio is not still successful, this experiment were designed for establishing a tissue culture techniques of pistachio. Shoots collected from grafted tree (7yrs)of pistachio were used as explant in primary culture with different treatments of medium(MS, modified MS,Q-L,DKW), plant hormone(BA,IBA,NAA,GA), disinfectant(7%NaClO3, 0.1%mercuric chloride), addition(folic acid, calcium D-pantothenate and LH), glucose and sucrose, experiment results shown that explant of pistachio were disinfected well with 0.1% mercuric chloride for 10min, the suitable primary medium for pistachio was: 1/2DKW+6-0.5mg/l 6-BA +0.5mg/l IBA +3.0 mg/l GA3 +1mg/l folic acid+ 300mg/l LH +1mg/l calcium D-pantothenate+15g/l glucose+6.5g/l agar.
