不同冻精处理方法和添加咖啡因对牛体外受精的影响

Effects of Different Treatment Methods of Frozen Semen and Addition of Caffeine on IVF in Cattle

摘要: 【目的】为了探讨不同冻精处理方法以及添加精子活力激活剂并孵育不同时间获能对体外受精及受精卵发育的影响。【方法】研究对体外受精前的牛冷冻精液分别采用直接离心法、上游法和Percoll法3种方法处理,并对处理后的精液分别用含2.5mM或5.0mM咖啡因的获能液(BO液+10μg/ml肝素钠)在培养箱内预先孵育10min或30min进行获能,不同处理的精子经体外受精后。【结果】结果表明:(1)3种冻精处理方法(直接离心洗涤法、上游法、Percoll法)对精子处理获能,IVF后受精卵的卵裂率(依次为51.58%、52.83%、53.20%)均无显著差异(P>0.05);桑椹胚、囊胚发育率Percoll法处理组(16.66%、10.18%)与上游法处理组(17.85%、16.07%)以及Percoll法处理组与直接离心洗涤法组(8.77%、7.01%)均无统计学差异(P>0.05),同时实验结果还表现出上游法处理组比Percoll处理组的桑囊胚率有所提高,且上游法处理组与洗涤法处理组在桑、囊率上出现了显著差异(P<0.05)。(2)获能液中添加咖啡因组比非添加组(对照组)卵裂率和桑囊胚率有所增加,其中添加5.0mM咖啡因组精子孵育30min的获能效果最佳,卵裂率、桑囊率达到68.59%、28.50%。【结论】以上结果表明上游法处理牛冷冻精液稍优,更有利于以后的受精卵发育;添加咖啡因能提高精子活力,且高浓度的咖啡因维持精子高活力持续时间较长。

Abstract: 【Objective】 In order to understanding effects of different treatment methods of frozen semen and addition of caffeine on IVF in bovine, 【Methods】 frozen-semen of bovine was treated by using Swimming-up method, Percoll method and Washing method, capacitated by adding 2.5mM or 5.0mM of caffeine in capacitation medium(BO medium+10μg/ml heparin sodium) and pre-incubated 10min or 30min to observe respective effect on the in vitro development ability of fertilized eggs. 【Results】 The results showed that (1)there was not any significant differences among cleavage rate(51.58%,52.83%,53.20%)(P>0.05) of fertilized eggs treated by frozen-semen from Washing method ,Swimming-up method and Percoll method,there was also not any significant differences on the in vitro development rate of morula or blastocyst between treatment of Percoll method(16.66%,10.18%) and Swimming-up method(17.85%,16.07%),and between treatment of Percoll method and of Washing method(8.77%, 7.01%)(p>0.05).But it can be seen from experimental results that in vitro development of morula and blastocyst of treatment of Swimming-up method had some enhance compared with that of treatment of Percoll method, there was significant differences of in vitro development rate of morula or blastocyst between treatment of Swimming-up method and of Washing method(P<0.05).(2)Addition of caffeine in fertilization medium can promote activity of sperm. Cleavage rate or development rate of morula or blastocyst in the presence of caffeine were higher than that of in the absence of caffeine, there was best effect of capacitation on sperm(Cleavage rate is 68.59%,blastocyst rate is 28.50%).【Conclusion】This indicated that, compared these 3 methods, Swimming-up method perhaps is economical and simple method of treatment methods of frozen-semen, it can be applied for IVP of embryo of bovine; This also showed that higher concentration of caffeine can maintain strong activity of sperm at comparative long period of time.
