香蕉枯萎病菌细胞壁降解酶的诱导及其对香蕉组织的降解

Induction of extracellular cell wall-degrading enzymes from Fusarium oxysporum f.sp. cubense and their effect on degradation of banana tissue

摘要:

本研究分别以5 种不同碳源诱导香蕉枯萎病菌4 号小种(Fusarium oxysporum f.sp. cubense race4,FOC-R4)产生细胞壁降解酶(CWDEs)。结果表明,以麸皮和果胶为碳源,可在活体外诱导FOC-R4高产纤维素酶(包括内切葡萄糖苷酶和β-D-葡萄糖苷酶)和果胶酶(多聚半乳糖醛酸酶);测定表明这两种酶对不同香蕉品种假茎组织均具有降解破坏作用,其中果胶酶的降解能力比纤维素酶的强。感病香蕉品种经果胶酶液处理24 h后释放的还原糖量约为抗病品种释放的1.82倍,反映了不同抗、感香蕉品种对FOC-R4的敏感性和抵御能力差异。

Abstract:

Five different carbon sources were used to study the induction of extracellular cell wall-degrading enzymes (CWDEs) from Fusarium oxysporum f.sp. cubense race4 (FOC-R4). The results showed that wheat bran and pectin can effectively induce cellulase (endo-1, 4-β-D-glucanase and β-D-glucosidase) and pectinase (polygalacturonase) from FOC-R4 respectively in vitro. It is indicated that both cellulase and pectinase can degrade pseudostem tissues of different cultivars of Musa spp., and the degradability of the latter was stronger than that of the former. After being treated for 24 hours with crude pectinase solution, reducing sugar content released by susceptible cultivars was about 1.82 folds of resistant ones, there were differences in sensitivity and resistance between resistant and susceptible cultivars to FOC-R4.
