ITS序列在西红柿种质资源鉴定中的应用研究

Application Research of ITS Sequences in Tomato Germplasm Identification

摘要:

本研究根据番茄的45S rDNA序列设计引物,以西红柿基因组DNA为模板通过PCR方法扩增出ITS(internal translation spacer)序列,在克隆测序的基础上对番茄属两个野生种与三个栽培种进行分子鉴定与亲缘关系分析。结果表明:(1)五个供试材料ITS序列的变异小,GC含量高,为57.0%~65.4%。(2)用不同的聚类方法分析发现三个栽培种供试材料亲缘关系较近,而野生种醋栗番茄(Solanum pimpinellifolium)都处于不同的分支类群,表明其与其他品种亲缘关系相对较远,同时发现其生物学特性与其他品种明显不同。

Abstract:

In this study, the primers of sequence was designed on tomato 45S rDNA , tomato genomic DNA as a template was amplified by PCR out of ITS sequences (internal translation spacer) , the cloned sequence is based on two wild tomato species and three cultivated species for molecular Identification and molecular analysis of genetic relationship. The results showed that: (1) five of the test materials are a small variation of ITS sequences, GC content was high up to57.0% ~ 65.4%. (2)using different clustering analysis showed that three cultivars tested materials have a closed genetic relationship, while the wild species of currant tomato (Solanum pimpinellifolium )are in different branches of taxa, indicated that their kinship with other species is relatively distant, at the same time the biological characteristics of Solanum pimpinellifolium significantly are different from other species.
