Viscoelastic Changes and Cold Tolerance Comparison of Arabidopsis and Tobacco Cells Under Low Temperature Stress

Abstract: Positively charged poly(diallyldimethylammonium chloride) (PDADMAC)modified quartz crystal microbalance (QCM) was used for the real-time monitoring of the viscoelastic changes of Arabidopsis suspension cells and BY-2 cells when they were subjected to low temperature stress to compare cold tolerance of these two types of plant cells. The results showed that as the decrease of the temperature, the cell viscoelastic index (CVI) of both types of the cells decreased and the cells became harder, the lower the temperature, the larger the drop in CVI. Over the temperature drop range (25°C-5°C), the change in CVI for Arabidopsis cells is smaller than that of BY-2 cells, which is consistent with that Arabidopsis has better cold tolerance than that of Nicotiana tabacumL. Therefore, we propose to use CVI as a new parameter to indicate the cold tolerance of plant cells.
