The article analyzes the mechanism of browning in tissue-culture on Amorphohphallus Konjac C. Koch from its generant reason, qualification and phenomenon. And concludes the preventing measures according to the fresh progress in recent years: choosing the young and the big explants, gathering the explants should avoid the time which the temperature is high, advance-cultivating the explants; the concentration of 2，4-D is proper low and the CTK is related high; Putting the culture medium under the low temperature and dark condition; Immerging the cut-surface in the culture medium, transforming the first inoculated generation explants into fresh culture medium as soon as possible ; respectively adding the active carbon(500mg/L), cysteine, sulfur dioxide,（100mg/L）PVP or Vc into the medium.
