Porcine Circovirus Type II Infection Inhibits the Humoral Immune Response to Pseudorabies Virus Vaccine

摘要: 为了探讨猪圆环病毒II型(PCV2)感染对伪狂犬病病毒(PRV)疫苗体液免疫反应的影响,检测了PCV2实验感染仔猪血清中PRV疫苗抗体含量的动态变化。将仔猪随机分成二组,一组仔猪进行PCV2接种和PRV疫苗免疫而作为PCV2组;另一组仔猪不予PCV2接种但进行PRV疫苗免疫而作为对照组。所有仔猪在PCV2接种后14天均进行PRV弱毒疫苗免疫,用间接ELISA检测仔猪血清中病毒特异性抗体;到PCV2接种后14天,PCV2组仔猪全部出现PCV2病毒血症。在PRV疫苗免疫后7天,多数仔猪血清中可检测到疫苗抗体之后疫苗抗体含量逐渐上升,在免疫后14天时对照组疫苗抗体水平显著(P<0.05)高于PCV2组,但在免疫后21天时两组之间抗体水平没有差异。研究结果显示:PCV2实验感染仔猪的PRV疫苗抗体产生出现延迟,表明PCV2感染可抑制PRV疫苗诱导的体液免疫反应。

Abstract: To explore the effect of the infection of Porcine Circovirus Type II (PCV2) on the humoral immune response to the pseudorabies virus (PRV) vaccine, the profile of the antibodies against the PRV vaccine in sera of the piglets infected experimentally with PCV2 were determined. The piglets were randomly divided into two groups. The piglets inoculated with PCV2 and vaccinated against PRV were served as the PCV2 group, and the piglets not inoculated with PCV2 but vaccinated against PRV were served as the control. All the piglets were vaccinated against the attenuated PRV vaccine at 14 days post inoculation (DPI) of the PCV2. The specific antibodies to the viruses in piglet sera were detected with indirect ELISA. By 14 DPI, all piglets in the PCV2 group had developed the viremia to PCV2. At 7 days post vaccination (DPV) against the PRV, the antibodies specific to the vaccine could be determined in most piglet sera, thereafter, the concentration of the vaccine antibodies increased gradually. At 14 DPV, the level of the vaccine antibodies in the control group piglets was significantly (P<0.05) higher than those in the PCV2 group. At 21 DPV, however, there were no significant differences in the vaccine antibodies level between the two groups. These data showed that the production of the antibodies specific to the PRV vaccine in the experimental PCV2-infected piglets was delayed, and indicated that the PCV2 infection played an inhibitory role on the humoral immune response to the PRV vaccine.
