南方根结线虫延长因子2基因cDNA全长克隆和序列分析

Cloning and Sequence Analysis of Full-length cDNA of Elongation Factor 2 Gene of Meloidogyn incognita

摘要:

以南方根结线虫为材料,根据同源序列法分别进行3′末端和5′末端的RACE,获得延长因子基因2 cDNA全长。此cDNA全长序列为2434 bp,包括2334 bp的完整ORF,编码一含777个氨基酸的多肽。该基因命名为Mi-eft2,其推导蛋白MI-EFT2的编码氨基酸与秀丽隐杆线虫的Ce-eft1,Ce-eft2,Ce-eft3,Ce-eft4基因的推导蛋白的同源性分别为26.7%、86.9%、13.8%和8.9%。蛋白功能分类预测MI-EFT2是一种在蛋白质延长过程中起作用的GTP水解酶,与真核生物的延长因子2功能相同。

Abstract:

The full-length cDNA sequence of elongation factor 2 (Mi-eft2) was obtained by RT-PCR and RACE from Meloidogyne incognita. It consisted of 2434 nucleotides, and encoded 777 amino acids. The amino acid sequence of deduced protein MI-EFT2 was 26.7%, 86.9 %, 13.8% and 8.9% identical with the deduced proteins of Ce-eft1, Ce-eft2, Ce-eft3 and Ce-eft4 from Caenorhabditis elegans respectively. Protein function prediction of MI-EFT2 suggested that it was a GTPase just like the eukaryotic elongation factor EF2, involved in the protein elongation process.
