小麦花粉多糖对微生物的抑制作用研究

摘要: 为研究小麦花粉多糖对微生物的抑制作用,采用温差破壁法和酶解破壁法相结合的方法对小麦花粉进行破壁处理,从中提取活性多糖,并利用甲醇分级沉淀法进行纯化。实验采用两因素随机区组设计,并对小麦花粉多糖的抑菌作用进行方差分析和多重比较检验。结果表明,小麦花粉多糖对细菌具有极显著的抑制作用,对真菌的抑制效果也比较显著。其中,以35%甲醇浓度,3000 r/min转数进行离心分离的多糖对细菌和真菌的抑制效果最佳;以30%甲醇浓度,4000r/min离心和以30%甲醇浓度,3500r/min离心的纯化工艺得到的活性多糖对细菌的抑制效果比较显著;采用其他甲醇浓度,离心转数的纯化工艺得到的活性多糖对细菌的抑制效果一般。

Abstract: Wheat Pollen polysaccharides on inhibition of microbial. Wheat pollen was carried out with cell-wall-broken treatment through temperature difference wall-broking method and enzymolysis wall-broking method in this study. Active polysaccharide was extracted from cell-wall-broken pollen and was purified by use of methyl alcohol fractionated precipitation method. Experiments using two-factor randomized block design, and the inhibitory effect of the polysaccharides of wheat pollen analysis of variance and multiple comparison tests. The results indicated that Wheat pollen polysaccharide had very significant inhibitory effects to bacteria and had significant inhibitory effects to fungi. The polysaccharide purified by 35% of methyl alcohol with 3000r/min centrifugation had the best inhibitory effect on bacteria and fungi. Methanol concentration of 30%, 4000r/min centrifuge and the inhibitory effect of active polysaccharides from the bacteria to 30% methanol concentration, 3500r/min centrifugal purification process significantly; other methanol concentration, obtained by the centrifugal rotation of the purification process activity polysaccharides on the bacterial inhibition effect.
