巴西橡胶树HbSIP1基因启动子的克隆和序列分析

Cloning and Sequence Analysis of HbSIP1 Promoter from Hevea brasiliensis

摘要: 为研究橡胶焦磷酸酶基因的表达特征及其在天然橡胶生物合成中调控机理,根据巴西橡胶树焦磷酸酶基因HbSIP1序列,利用GenomeWalker方法对HbSIP1基因启动子序列进行了分离,对其序列进行了生物信息学分析,并构建了含有该序列的植物表达载体pSIP1-1381Z。结果表明:分离获得了1198 bp的HbSIP1基因启动子序列,该序列具有真核生物典型启动子结构特征,并含有众多应答激素和胁迫信号的调控元件。对HbSIP1的启动子区的克隆和分析,为进一步研究HbSIP1的功能和表达调控机制奠定基础。

Abstract: The aim was to study the expression characteristics and regulation mechanism of pyrophosphatase gene in natural rubber biosynthesis. Based on the sequence of pyrophosphatase gene HbSIP1 from H. brasiliensis, the promoter sequence of HbSIP1 was cloned from H. brasiliensis by GenomeWalker method, the sequence was analyzed and a plant expression vector with the promoter sequence of HbSIP1 was constructed. Sequence analysis showed that the 1198 bp promoter sequence was obtained and its sequence had typical promoter features of eukaryote and contained several regulatory elements related to hormone and stress responses. Cloning and analysis of the promoter region of HbSIP1 could lay foundation for the further study of the function and expression regulation mechanisms of HbSIP1.
