基于流式细胞术的澳洲坚果基因组C值测定

Estimation of Genomic C Value of Macadamia integrifolia spp. by Flow Cytometry

摘要: 为建立澳洲坚果基因组C值的流式细胞术测定方法,采用改良的OTTO细胞核提取液配方和PI荧光染料,以‘O.C’嫩叶为材料,以基因组DNA含量已知的Pisum sativum L. Ctirad和Zea mays L. CE-777为对照,运用流式细胞术对澳洲坚果基因组C值进行了测定。结果表明,澳洲坚果‘O.C’二倍体基因组DNA总量为1.91488599 pg,其C值为0.957443 pg;按1 pg DNA=0.978×109 bp来计算,‘O.C’基因组大小为1.872758×109 bp;采用2种不同对照材料作为内标测定的结果无显著差异。本研究发现,运用流式细胞术进行澳洲坚果基因组C值测定准确可靠,可为其他物种基因组C值测定提供参考。

Abstract: In order to establish the flow cytometry (FCM) method for determination of macadamia genomic C value, The tender leaves of macadamia cultivar ‘O.C’ was used as material, the adjusted OTTO and PI were used as isolation buffer and DNA fluorochrome, the Pisum sativum L. Ctirad and Zea mays L. CE-777 were used as DNA reference standards, the genomic C value was determined by FCM. The result showed that the DNA content of diploid macadamia spp. was 1.91488599 pg and the genomic C value was 0.957443 pg; the genome size of macadamia spp. was 1.872758×109 bp calculated by 1pgDNA=0.978×109 bp. The results determined by two different DNA reference standards have no significant difference. The study found that the FCM method was accurate and reliable, it provided a valuable reference for genomic C value determination of other species.
