地鳖提取物制备和体外抗氧化活性的研究

Preparation of Eupolyphaga Sinensis Walker Extracts and Their Antioxidant Activity in Vitro

摘要: 为研究地鳖不同提取物的体外抗氧化活性。分别通过蛋白酶水解、水提法制备地鳖多肽、多糖和水提物,采用邻苯三酚自氧化、Fe2+-邻二氮菲法和分光光度法分别检测地鳖不同提取物对超氧阴离子自由基(O2-·)、羟自由基(OH·)和二苯代苦味酰基自由基(DPPH·)的清除,分光光度法分别检测红细胞氧化溶血和肝脏线粒体肿胀吸光度值,硫代巴比妥酸(TBA)法测定小鼠不同组织丙二醛(MDA)含量。地鳖多肽、地鳖多糖,地鳖水提物组对O2-·、OH·和DPPH·三种自由基的清除率显著低于阳性对照组,随着提取物浓度的升高对自由基清除率增加;随着地鳖三种提取物浓度提高红细胞溶血吸收值比溶血组显著降低,地鳖水提物组红细胞溶血率明显低于地鳖多肽和多糖组。地鳖多肽组肝脏MDA生成抑制率最高,地鳖多糖组肾脏MDA抑制率高于地鳖多肽和水提物组,随着地鳖提取物浓度提高各组织中MDA生成抑制率升高;随着反应时间延长地鳖提取物组肝脏线粒体吸光度值高于肿胀对照组。地鳖多肽、多糖和水提物有效的清除三种自由基,抑制红细胞溶血和肝脏线粒体肿胀,保护肝肾组织氧化损伤,具有一定程度体外抗氧化活性。

Abstract: This paper was to investigate the antioxidant activity of different kinds of extracts from Eupolyphaga sinensis Walker (ESW) in vitro. Polypeptides, polysaccharides and water extracts from ESW were prepared by protease hydrolysis and water extracting methods. The scavenging capacity of different kinds of ESW extracts on superoxide anion, hydroxyl and 1,1-Diphenyl-2-2picrylhydrazyl (DPPH·) free radicals were detected by pyrogallol autoxidation, Fe2 -phenanthroline method and spectrophotometry methods, respectively. Spectrophotometry was used to assay absorbance values of the hemolysis of red blood cells and liver mitochondrial swelling in the mice. Malondialdehyde (MDA) contents of different tissues from the mice were detected by Thiobarbituricacid (TBA). There was a significant decrease in the scavenging capacity of polypeptides, polysaccharides and water extracts from ESW on superoxide anion, hydroxyl and DPPH free radicals compared with the positive control group, and an increase in the scavenging rate with the extract concentration increasing. As the concentrations of three kinds extracts from ESW were increasing, hemolysis absorption values of red blood cells in experimental groups were more markedly decreased than that in the hemolysis group, while there was a significantly lower hemolysis rate in water extract group compared with polypeptides and polysaccharides groups. MDA inhibition rate of the liver in the polypeptide group was the highest, and that of the kidney in polysaccharide group was higher than in polypeptide and water extract groups. There was an increase in MDA inhibition rate with the increasing of three kinds of ESW extracts. The absorbance value of the liver mitochondria in ESW extract groups was higher than that in the control group with prolong of reaction time. The results showed that polypeptides, polysaccharides and water extracts from ESW had an effective scavenging capacity on three kinds of radicals, an inhibition in hemolysis of red blood cells and hepatic mitochondrial swelling, and a protection of liver and kidney tissues from oxidant injury, which suggested ESW extracts had antioxidant activity in vitro to a certain degree.
