芦竹高效快繁体系的建立

摘要: 研究旨在建立芦竹(Arundo donax)高效组培快繁体系,为其大规模工厂化生产、应用及再生遗传转化体系的建立提供基础。以芦竹腋芽茎段为外植体,分别用不同激素进行正交实验,对芦竹的腋芽萌发、增殖及生根培养基进行筛选和优化。实验发现,MS+ 4.00 mg/L 6-BA+ 1.00 mg/L IBA为芦竹腋芽萌发最佳培养基配方,萌发率为96.7%;MS+ 7.50 mg/L 6-BA+ 1.00 mg/L IBA为最佳增殖培养基配方,增殖系数可达13.5,单株增殖系数可达16,其增殖系数较之前的研究提高了1倍多;1/2MS+ 1.00 mg/L IBA,为最佳生根培养基配方,生根率最高,为93.8%,平均根数6.33条;移栽培养选择高7 cm以上,根长1~3 cm的健壮组培苗进行驯化移栽效果最好。本研究成功建立了芦竹完善的高效组培快繁体系,有效提高了芦竹的繁殖系数,同时为芦竹的再生及分子水平的育种提供基础。

Abstract: The study is to find out the most efficient way for rapid propagation of Arundo donax,which can lay the foundation for its industrial production, application and transformation. Stems with auxiliary buds were used as explants, different concentrations of plant growth regulators were tested to find the most effective protocol for rapid propagation of Arundo donax. MS 4.00 mg/L 6-BA 1.00 mg/L IBA was the most efficient initial medium for the sprouting of auxiliary buds, the germination rate could get to 96.7%; MS 7.50 mg/L 6-BA 1.00 mg/L IBA was the most efficient proliferation medium, and the average proliferation rate was 13.5, while individual proliferation rate reached 16; 1/2MS 1.00 mg/L IBA was the most suitable rooting medium, in which the rate of rooting was 93.8%, the average root number was 6.33. Plants, which were 7 cm high and with roots 1 cm to 3 cm long, were chosen for further acclimatization. The research establishes the rapid propagation of Arundo donax, improves the propagation coefficient and provides evidence for transformation and breeding at molecular level.
