‘嘎啦’苹果试管苗生根培养基的筛选及SA对试管苗移栽的生理效应

Rooting Cultivation Medium for‘Gala’Apple Plantlet and Physiological Effect of SA on Transplanting

摘要: 苹果试管苗生根培养及移栽困难、成活率低,为筛选适宜的生根培养基及移栽的适宜外源物质,试验以继代培养的‘嘎啦’苹果茎段为材料,研究基本培养基及吲哚乙酸(IAA)、吲哚丁酸(IBA)和萘乙酸(NAA)3种生长素及水杨酸(SA)对生根及移栽成活的影响,结果表明:基本培养基、生长素种类及浓度显著影响试管苗的生根率,1/2MS显著优于MS培养基,IAA优先其他两种生长素;外源SA可以显著提高试管苗的移栽成活率,并存在浓度效应。适宜‘嘎啦’生根的培养基为1/2MS+0.2 mg.L-1 IAA,0.8mg.L-1 SA处理可显著降低气孔开度、提高叶片叶绿素含量而显著提高移栽成活率。

Abstract: Because rooting cultivation and transplanting of apple test- tube plantlets is difficult, and the survival rate is low, in this study,‘Gala’apple test-tube plantlets were taken as experiment materials, to investigate the effect of basic culture medium and three kinds of auxin and salicylic acid (SA), including indole-3-acetic acid (IAA), indole-3-butyric acid (IBA) and 1-naphthylacetic acid (NAA), on rooting and transplanting survival rate. The results showed that the basic medium, hormone type and concentration obviously influenced the rooting percentage of tube seedlings, 1/2MS was significantly better than the MS culture medium, IAA was superior to the other two kinds of auxins; exogenous SA could significantly increase the transplanting survival rate of tube seedlings and had the concentration effect. Appropriate‘Gala’rooting medium was 1/2MS IAA 0.2 mg/L, 0.8 mg/L salicylic acid treatment could significantly reduce the stomatal opening and significantly improve the survival rate of transplanting.
