β-巯基乙醇对不同时期芒果剪口芽总RNA 质量的影响

Effect of β-mercaptoethanol on Total RNA Quantity of Mango Pruning Wound Buds in Different Periods

摘要: 芒果剪口芽中富含多酚,由于多酚易被氧化,且难以与核酸分离,导致总RNA提取困难。为提取适于芒果实时荧光定量PCR分析的高质量RNA,通过在华越洋超快新型植物RNA提取试剂盒的细胞裂解液中加入适量的β-巯基乙醇,研究β-巯基乙醇对芒果剪口芽总RNA提取质量的影响。试验结果表明,在细胞裂解液中加入300 μL β-巯基乙醇,可以排除多酚、色素等杂质的干扰,获得条带完整、质量较好的总RNA样品。RT-PCR实验结果进一步表明,此方法提取的RNA完全适于后续分子生物学试验研究。

Abstract: It is difficult to extract total RNA from the pruning wound buds of mango since those buds are rich in phenols which are easily oxidized and hardly separated from the nucleic acid. In order to extract high quality RNA which was suitable for real-time fluorescent quantitative PCR. The influence of β-mercaptoethanol on the quality of RNA extraction was discussed by adding an appropriate amount of β-mercaptoethanol into the cell lysis buffers of quick RNA isolation kit which was made by HUAYUEYANG BIOTECHNOLOGY (BEIJING) CO. LTD. The results showed that adding about 300 μl β-mercaptoethanol into the cell lysis buffers, which could eliminate the interference impurities such as phenols, pigments, et al. Total RNA isolated by this method was of high quality with a full strip, and the result of RT-PCR indicated that it was able to be used for later molecular manipulations.
