湖北省不同克氏原螯虾群体携带白斑综合征病毒情况调查

Investigation of White Spot Syndrome Virus in Different Procambarus clarkii Populations in Hubei

摘要: [目的]为了了解湖北省克氏原螯虾的白斑综合征病毒(white spot syndrome virus, WSSV)携带情况。[方法]2016年5月—2017年7月对湖北省几个养殖地区采集的741个克氏原螯虾样品进行了WSSV的PCR检测,并选取了49个PCR阳性扩增产物进行测序分析。[结果]结果显示,湖北省潜江市、监利县等主要养殖区的克氏原螯虾和洪湖市新堤地区克氏原螯虾群体携带WSSV病毒的比率为65%以上;鄂州市、随州市以及洪湖市螺山镇养殖的克氏原螯虾群体携带WSSV病毒的比率为40%左右。测序结果发现,有部分样品中WSSV的保守序列发生碱基A/T的突变。系统发育树分析结果显示,发生碱基A/T突变的WSSV与17个已公布的WSSV地区亚种的亲缘关系都比较远,而未发生A/T碱基突变的WSSV与澳大利亚株(MF161441)、孟加拉株(KJ719075)亲缘关系更近。[结论]研究结果可为克氏原螯虾WSSV的防控提供一定的参考。

Abstract: To obtain the white spot syndrome virus (WSSV) carrying situation in Procambarus clarkii, PCR detection on WSSV was conducted on 741 samples of Procambarus clarkii from 7 aquaculture areas in Hubei from May 2016 to July 2017. 49 positive PCR products were sequenced and analyzed. The results showed that over 65% of the samples carried WSSV from areas of Qianjiang City, Jianli County and Xindi in Honghu City, and about 40% of the samples carried WSSV from areas of Ezhou City, Suizhou City and Luoshan Town in Honghu City. The sequencing results indicated that A/T base mutation was found in the conserved region of WSSV of some samples. The phylogenetic tree analysis displayed that there was a farther genetic relationship between the WSSV samples with A/T base mutation and 17 WSSV subspecies in GenBank. However, a closer genetic relationship was found between the WSSV samples without A/T base mutation and the strains from Australia (MF161441) and Bangladesh (KJ719075). This study could provide a reference for the prevention and control of WSSV in Procambarus clarkii.
