7种外生菌根真菌的生长曲线测定研究

Seven Ectomycorrhizal Fungi: Determination of the Growth Curve Characteristics

摘要:

为了解外生菌根真菌在实验室培养条件下的生长特性,选取灰色锁瑚菌[Clavulina cinerea (Bull.) J. Schrōt.]、绒白乳菇[Lactarius vellereus (Fr.) Fr.]、白乳菇[Lactarius piperatus (L.) Pers.]、网纹马勃[Lycoperdon perlatum Pers.]、赭褐马勃[Lycoperdon umbrinum Pers.]、光盖松塔牛肝菌[Strobilomyces glubricaps W.F. Chiu]和点柄乳牛肝菌[Suillus granulatus (L.) Roussel]为研究对象,采用固体PDA培养基培养、十字交叉法测定菌落生长直径与改良液体PD培养基培养测定菌丝生物量结合的方法测定7种外生菌根真菌的生长曲线。结果表明,不同外生菌根真菌在固体培养基培养条件下生长速率存在明显差异,具体表现出生长速率:光盖松塔牛肝菌>灰色锁瑚菌>绒白乳菇>赭褐马勃>网纹马勃>点柄乳牛肝菌>白乳菇;不同外生菌根真菌在液体培养基培养条件下生长速率存在明显差异,具体表现出:灰色锁瑚菌>光盖松塔牛肝菌>网纹马勃>绒白乳菇>赭褐马勃>白乳菇>点柄乳牛肝菌。综上,同种外生菌根真菌在固体和液体两种培养基培养条件下的生长速率并不完全一致。在生产上,单凭菌落的大小无法反映菌种真实的生长状况。

Abstract:

In order to understand the growth characteristics of ectomycorrhizal fungi under laboratory culture conditions, Clavulina cinerea (Bull.) J. Schrōt., Lactarius vellereus (Fr.) Fr., Lactarius piperatus (L.) Pers., Lycoperdon perlatum Pers., Lycoperdon umbrinum Pers., Strobilomyces glubricaps Chiu and Suillus granulatus (L.) Roussel were selected as the materials. The growth curves of the 7 ectomycorrhizal fungi were determined by the combination of solid PDA medium, cross method (measuring the colony growth diameter ) and the improved liquid PD medium (measuring the biomass of hyphae). The results showed that there were significant differences in the growth rates of different ectomycorrhizal fungi under the condition of solid medium culture, and the growth rates were as follows: Strobilomyces glubricaps W.F. Chiu > Clavulina cinerea (Bull.) J. Schrōt. > Lactarius vellereus (Fr.) Fr. > Lycoperdon umbrinum Pers. > Lycoperdon perlatum Pers. > Suillus granulatus (L.) Roussel > Lactarius piperatus (L.) Pers.. The growth rates of different ectomycorrhizal fungi in liquid medium were significantly different, which were in an order of Clavulina cinerea (Bull.) J. Schrōt. > Strobilomyces glubricaps W.F. Chiu > Lycoperdon perlatum Pers. > Lactarius vellereus (Fr.) Fr. > Lycoperdon umbrinum Pers. > Lactarius piperatus (L.) Pers. > Suillus granulatus (L.) Roussel. In conclusion, the growth rate of the same ectomycorrhizal fungi in solid and liquid media is not completely consistent. In production, the size of colonies alone cannot reflect the real growth status of strains.
