贝莱斯芽孢杆菌(Bacillus velezensis)YFB3-1高密度液体发酵条件优化

摘要:

旨在获得高密度的贝莱斯芽孢杆菌(Bacillus velezensis)YFB3-1生防菌剂,用于预防作物土传病害的发生。采用单因素和正交试验对菌株YFB3-1的液体培养基和培养条件进行优化。在酵母粉8 g/L、复合氨基酸10 g/L、氯化钠1 g/L、硫酸镁(MgSO4·7H2O) 1 g/L的液体培养基中培养菌株YFB3-1,初始pH 7.0、接种量2.5%、发酵温度30℃、转速180 r/min和培养时间48 h为发酵条件时,获得的菌体浓度为2.89×1010 cfu/mL,是菌株YFB3-1液体发酵的最优培养基和培养条件,比牛肉膏蛋白胨液体培养基(NB)的菌体浓度4.77×109 cfu/mL,提高了5.06倍,在相同时间内可以提高发酵效率。本研究为生防菌B. velezensis YFB3-1的产业化扩大培养和推广应用提供了理论依据和技术参数。

Abstract:

The aim is to obtain high density Bacillus velezensis YFB3-1 biocontrol agent which can be used to prevent soil-borne diseases of crops. In this study, single factor and orthogonal experiments were used to optimize the liquid medium and culture conditions of strain YFB3-1. The results showed that strain YFB3-1 was cultured in liquid medium with yeast powder 8 g/L, compound amino acid 10 g/L, sodium chloride 1 g/L and magnesium sulfate (MgSO4·7H2O) 1 g/L, and when the fermentation conditions were initial pH of 7.0, inoculation amount of 2.5%, fermentation temperature of 30℃, rotation speed of 180 r/min and culture time of 48 h, the bacterial concentration was 2.89×1010 cfu/mL, and the optimal culture medium and culture conditions were obtained for the liquid fermentation of strain YFB3-1. Compared with beef extract peptone liquid medium (NB), in which the bacterial concentration was 4.77×109 cfu/mL, the bacterial concentration under the fermentation conditions of this experiment increased by 5.06 times, and the fermentation efficiency could be improved in the same time. The study provides a theoretical basis and technical parameters for the industrial expansion and application of B. velezensis YFB3-1.
