利用简便SSR法鉴定‘兴蔬早佳’丝瓜杂交种纯度

Rapid Identification of ‘Xingshu Zaojia’ Luffa Hybrid Purity by SSR Molecular Markers

摘要:

为保障‘兴蔬早佳’丝瓜杂交种的纯度,需建立一套安全可靠、简单快速的杂交种纯度鉴定方法。本研究利用简化的SSR分子标记技术,以‘兴蔬早佳’丝瓜品种杂交种及其父母本为试材,从50对SSR引物中筛选出1对能稳定扩增杂交种与其亲本,且扩增产物呈共显性的引物。利用该引物对不同地点生产的‘兴蔬早佳’杂交种进行纯度鉴定,结果与田间表型鉴定结果误差小于1.2%。研究获得1对适于鉴定‘兴蔬早佳’杂交种纯度的引物,建立了一套适于该品种种子纯度鉴定的简便方法。利用该引物对‘兴蔬早佳’丝瓜杂交种种子纯度进行鉴定,不仅节约了鉴定成本,更缩短了鉴定周期,可为该品种商品种的安全销售提供技术保障。

Abstract:

In order to ensure the purity of ‘Xingshu Zaojia’ luffa, a set of safe, reliable, simple and fast hybrid purity identification methods should be established. In this research, we adopted the simplified SSR molecular marker technology, used the hybrid of ‘Xingshu Zaojia’ luffa and its parents as test materials, and screened a pair of primers that could stably amplify the hybrid and its parents from 50 pairs of SSR primers, and the amplification products were co-dominant. This pair of primers was used to identify the purity of ‘Xingshu Zaojia’ hybrids produced in different places, and the error between the simplified SSR and the field morphological identification results was less than 1.2%. A pair of primers suitable for identifying the purity of ‘Xingshu Zaojia’ hybrid was obtained, and a set of simple methods suitable for identifying the seed purity of this variety was established. Using this pair of primers to identify the seed purity of ‘Xingshu Zaojia’ luffa hybrid can not only save the identification cost, but also shorten the identification cycle, which can provide technical guarantee for the safe sale of the commercial variety.
