基于响应面法优化环烯醚萜苷提取工艺研究

摘要:

通过响应面法优化秦艽药材中环烯醚萜苷类成分的提取工艺。在单因素试验的基础上,根据响应面法设计,以马钱苷酸、獐牙菜苦苷和龙胆苦苷3种环烯醚萜苷总含量作为考察指标,优化最佳提取工艺条件。研究得出最佳提取工艺条件,即以80%甲醇作为溶剂,温度50℃,超声功率720 W,液料比40 mL:1 g,提取时间30 min并连续提取2次。按照此条件,实际测定值和预测值分别为15.562%和15.904%,两者相差2.20%,其结果基本一致。本法操作工艺相对简单,检测结果比较理想,可以为秦艽药材中环烯醚萜苷类的提取工艺提供参考。

Abstract:

The extraction process of iridoid glycosides from Gentianae macrophyllae radix was optimized by response surface methodology. On the basis of single factor test, according to the design of response surface method, the total content of loganic acid, swertiamarin and gentiopicroside was used as the index to optimize the extraction conditions. The optimum extraction conditions were as follows: 80% methanol was solvent, extraction temperature was 50℃, ultrasonic power was 720 W, liquid-solid ratio was 40 mL: 1 g, extraction time was 30 minutes and extracted 2 times. According to this condition, the actual measured value and predicted value were 15.562% and 15.904%, with the relative deviation of 2.20%, and the results were basically the same. The operation process is relatively simple and the test results are ideal, which provides a reference for the extraction process of iridoid glycosides in Gentianae macrophyllae radix.
